Specificity
This antibody binds morphine and cross reacts with other opioids like opium, codeine and heroin. Morphine is a strong opiate that is found naturally in opium, a dark brown resin in poppies (Papaver somniferum). It is considered the classic opioid analgesic, most commonly used in pain management, morphine provides major relief to patients afflicted with pain. It is a non-synthetic narcotic with a high potential for abuse.

The original antibody was generated by immunizing BALB/c mice with morphine-BSA conjugated fusion protein. A mouse scFv library was generated from the spleen cells and the particular antibody was isolated by panning against morphine-conjugated thyroglobulin (morphine-C-Tg).
Application Notes
The binding reactivity of this antibody for morphine conjugated thyroglobulin (morphine-C-Tg) was done using ELISA. The original scFv antibody binds morphine-C-Tg with a binding affinity of Kd= 0.126 nM. It was reported that this antibody does not cross react with thyroglobulin conjugated codeine-, cocaine-, or ketamine. However, this antibody is capable of cross reacting with free morphine, opium, codeine and heroin. The IC50 values of M86 scFv for opium, morphine, codeine and heroin was 257 ng/mL, 36.4, 7.3 and 7.4 nM, respectively (Matsukizono et al., 2013; JP2012232948).
