Specificity
This antibody is specific for human VAP-1, a glycosylated homodimeric membrane protein consisting of two 90 kDa subunits connected by disulfide bonds. The recognized epitope is present both in dimeric and monomeric forms of VAP-1.

The original antibody was generated by immunizing specific pathogen-free BALB/c mice with affinity-purified VAP-1 obtained from tonsil stromal lysate.
Application Notes
The original version of this antibody (mouse IgG2a) was generated, and its specificity was characterized by reciprocal immunoprecipitation with another anti-VAP-1 antibody (1B2). In gel analyses, this antibody was shown to react with the dimeric 170-kDa VAP-1 and, under reducing conditions, with the monomeric 90-kDa form of VAP-1. Furthermore, it was used for both immunofluorescence staining and flow cytometry analysis to assess the expression of VAP-1 in rat HEV-like endothelial cell line Ax transfectants (Salmi et al., 1998; PMID: 9605169). The original version of this antibody was biotinylated and characterized via various assays. In an ELISA, it was used to detect serum VAP-1 levels. In a Western blot, it was employed to analyze VAP-1 presence and molecular weight. In immunoaffinity chromatography, it was used to purify VAP-1 from serum samples. In an adhesion assay, it was used to specifically deplete sVAP-1 from serum (Kurkijärvi et al., 1998; PMID: 9686623). This antibody was used in transwell migration assays to study the phenotype of transmigrated monocytes by flow cytometry; it was employed to pretreat primary human hepatic sinusoidal endothelial cells to study its effect on monocyte transmigration.
