Specificity
This antibody binds a conformational epitope on the human and mouse endosialin. Endosialin (also known as tumor endothelial cell marker molecule 1, TEM1/CD248) is a type I transmembrane protein, composed of a glycosylation modified 80.9KDa protein core region, the modified mature glycoprotein is about It is 175KDa. TEM1 is one of the human tumor markers, which plays an important role in tumor cell growth, angiogenesis, invasion and metastasis. Tumor endothelial cell marker molecule 1 (TEM1/CD248) is identified as a microvascular marker of tumor angiogenesis in various human cancers and highly expressed on a variety of solid tumors (endothelial cells and stromal cells), such as sarcoma, brain tumor, breast cancer, skin cancer, colon cancer, and experiments have shown that in ovarian cancer, The expression of TEM1 is closely related to tumor invasion and prognosis.

The original antibody was isolated from a yeast-display scFv library was constructed by homologous recombination of the thrombotic thrombocytopenic purpura (TTP) patient repertoire originally expressed on M13 bacteriophage in the novel vector pAGA2 for yeast-display expression. The library was panned against human TEM1 recombinant protein.
Application Notes
The in vitro binding characterization of this antibody was done using ELISA and flow cytometry. This antibody can recognize TEM1-expressed on mammalian cells, like human or murine cell lines expressing TEM1 endogenously (HEK-293T, MOV1 and 2H11) or stably transduced with pHRSINTEM1 (hTEM1-MS1, hTEM1-H5V and hTEM1-SKOV3) in a flow cytometric assay. This antibody binds specifically to Endosialin/TEM1-expressing ovarian tumor in vivo, with functional stability over 48 hrs. (PMID: 20837020). The original scFv version of this antibody binds human TEM-1 with a dissociation constant of Kd= 2 nM (PMID: 29313073).The scFv78-saporin immunoconjugate generated using this antibody exerted dose- dependent cytotoxicity with high specificity to TEM1-positive cells in vitro (PMID: 27933426). This antibody was used in the generation of a multivalent Fc-fusion protein called 78Fc, which has desirable characteristics for immuno-imaging applications.
