Key features and details | |
Cat. No. | MABL-852 |
Name | Anti-CD28 mAbs |
Clone No. | AFD- 1C6 |
From | Recombinant Antibody |
Isotype | Engineer antibody |
Application | activation, Blocking, FA, in vivo, ELISA, FC |
Species Reactivity | Dog |
Basic Information | |
Specificity | The antibody binds specifically to canine CD28, a self-surface receptor expressed by thymocytes and a subset of natural killer cells. CD28 acts as a receptor for CD80 and CD86, providing a costimulatory signal for the activation of T-cells and the production of interleukins, such as IL-6. |
Alternative Name | TP44; Costimulatory molecule B7 receptor CD28; T-cell costimulatory molecule CD28 |
UniProt | Q9N0N8 |
Immunogen | Canine CD28. |
Application Notes | This antibody exhibits an antagonistic activity, blocking mixed lymphocyte reactions and cytotoxic T lymphocyte-mediated killing. The use of this antibody to neutralize the CD26 signal following T-cell receptor activation is of clinical importance for transplantation therapy, as it has been shown to prevent graft rejection and graft-versus-host disease in the canine model of allogeneic hematopoietic cell transplantation (HCT). Interestingly, this antibody also exhibits agnostic activity; it was coated onto magnetic beads and used for the activation of canine peripheral blood lymphocytes (Szopa et al., 2021; PMID: 33679741). |

Note on publication | Describes the role of CD28 in the activation of T cells via costimulatory signals, and the blockade of this signal using anti-CD28 monoclonal antibodies such as 1C6. Also demonstrates the advantages of using this method in comparison to cytotoxic T lymphocyte antigen 4 (CTLA-4)-Ig fusion protein by in vivo studies in the dog HCT model. |
