Specificity | This antibody recongises human CD47 extracellular domain. CD47 is a a multitransmembrane glycoprotein widely expressed on all hematopoietic cells and most other cell types. It is involved in cell adhesion, Ca2+ influx, platelet activation and peripheral mononuclear neutrophils migration across endothelial and epithelial monolayers. |

Immunogen | This antibody was raised by immunising Balb/c mice with T84 epithelial membranes. |
Application Notes | C5/D5 is recommended for staining cell surface CD47 as well as neutralising CD47. Furthermore, it is capable of inhibiting peripheral mononuclear neutrophils migration across polarised monolayers of the human intestinal epithelial cell line T84 in a bidirectional fashion (Parkos et al., 1996). This antibody is also very effective in detecting CD47 in flow cytometry; this was demonstrated with various cells, such as CaCO2 and polymorphonuclear neutrophils (Liu et al., 2001). Finally, C5D5 was also utilised in the immunofluorescence staining of the basolateral surfaces of methanol-fixed CaCO2 monolayers expressing exogenously transfected human CD47 (Liu et al., 2001). |
