Application | SPR, ELISA |
Species Reactivity | Vibrio cholerae |
Basic Information | |
Specificity | This antibody binds the B subunit of the cholera toxin. |
Alternative Name | ctxB; toxB; VC_1456; Cholera enterotoxin subunit B; Cholera enterotoxin B chain; Cholera enterotoxin gamma chain; Choleragenoid |
UniProt | P01556 |
Immunogen | The original antibody was isolated from a non-immune semisynthetic llama library constructed from blood samples (6 ml) collected from the left jugular vein of three research naïve mature llamas. |
Application Notes | The binding specificity of this antibody towards cholera toxin was checked using ELISA (PMID:17165813). SPR was used to determine the dissociation constant (Kd) of VHH A9; the Kd was determined to be 77 ± 5 nM (PMID: 23385744). It was reported that two point mutations (N27D and R71I) help stabilize A9 wild-type form which has a relatively low melting point (PMID: 31342597). |
Antibody First Published | Goldman et al. Facile Generation of Heat Stable Antiviral and Antitoxin Single Domain Antibodies from a Semi-synthetic Llama Library. Anal Chem. 2006 Dec 15; 78(24): 8245–8255. PMID:17165813 |
Note on publication | A semisynthetic llama library was constructed from non-immune llamas and binders specific for live vaccinia virus (a smallpox virus surrogate), hen egg lysozyme, cholera toxin, ricin, and staphylococcal enterotoxin B were selected. |
