Specificity | This antibody binds Dxd, an anti-cancer alkaloid camptothecin analog, which is a potent DNA topoisomerase I (TOP1) inhibitor. By forming a stable ternary complex with TOP1 and DNA, it arrests the cell cycle to S phase, thereby preventing DNA replication and causing cell death. DxD is used for the generation of antibody-drug conjugates for targeted delivery of toxic materials to cancerous cells. Application Notes | The binding reactivity of this antibody towards Dxd-KLH and Dxd-BSA conjugates was determined using ELISA. This antibody is capable of binding antibody-drug conjugates ADC-8201 and ADC029 in a competitive ELISA, wherein the antibody was trastuzumab. The binding affinity of this antibody towards Dxd-BSA was determined using surface plasmon resonance (SPR) and it was reported to be Kd= 3.38nM (CN115677857). |
