Specificity | The antibody binds to the native EphA3 globular ephrin-binding domain. The antibody targets a site closely adjacent to the heterotetramerization site on the N-terminal of EPHA3's extracellular domain adjacent to the ligand-binding site and has a high affinity for EPHA3. |

Immunogen | The original antibody was prepared by intraperitoneal immunization of a BALB/c mouse with LK63 cells. |
Application Notes | This antibody was used for detection of hek expressed on LK63 cells by flow cytometry. Hek antigen was immunoprecipitated using the antibody from 125I-labeled LK63 cell lysates (Boyd et al., 1992; PMID: 1737782). The specificity of the chimeric version of the antibody was confirmed by ELISA analysis (US8664365). In a LK63 xenograft model, administration of the antibody inhibited the tumor growth, decreasing the spreading from the bone marrow to the spleen and other organs and increases the latency of the disease. Similar effects were observed in LK63 EPHA3 knock down xenografts. Further, antibody treatment of a xenograft model using EPHA3-transfected Reh, showed reduction in the bone marrow engraftment and increases the latency of the disease (Charmsaz et al., 2016; PMID: 27922598). The antibody bound to the native EphA3 globular ephrin-binding domain with subnanomolar affinity (KD ∼ 0.5 nM) (Smith et al.; 2004; PMID: 14660665). |
