Cat. No. | MABL-1549 |
Name | Anti-Factor VIII mAbs |
Clone No. | AFD- EL-14 |
From | Recombinant Antibody |
Isotype | Engineer antibody |
Application | functional assay, inhibit, IP |
Species Reactivity | Human |
Basic Information | |
Specificity | This antibody binds the C2 domain of the human factor VIII, a glycoprotein involved in blood coagulation. Factor VIII acts as a cofactor for factor IXa which converts factor X to the activated form Xa in the presence of calcium and phospholipids. Defects in the F8 gene, which encodes Factor VIII, result in the coagulation disorder hemophilia A. |
Alternative Name | AHF; FVIII; Factor 8; anti-hemophilic factor; Coagulation factor VIII; procoagulant component |
UniProt | P00451 |
Immunogen | The original antibody was isolated from a human phage display library generated from the peripheral blood of an inhibitor patient and panning the library against factor VIII light chain. |
Application Notes | The binding characterization of this antibody towards radiolabeled FVIII light chain and the C2 domain of factor VIII was done using immunoprecipitation. The inhibitory activity of this antibody against factor VIII was determined using a one-stage clotting assay. This antibody was capable of completely eliminating FVIII inhibition by CLB-CAg 117, a C2 domain directed anti-factor VIII antibody (PMID: 10627462). The scFv version of this antibody bound factor VIII light chain with a dissociation constant of Kd = 1.1 nM (PMID: 11929772). |
