Application Notes | A second-generation retroviral CAR, consisting of the scFv fragment of the antibody, with CD8 stalk, human CD3ζ and 4–1BB domains, targeting mouse FAP was developed. The Fap-CAR-T cells killed FAP-expressing 3T3 target cells specifically. The FAP-CAR-T cells were tested in vivo in three different established models, showing that the growth of tumors was significantly reduced by 35–50% following treatment. Further, the anti-FAP-CAR T cells demonstrated enhanced antitumor response when combined with a tumor vaccine. No significant toxicity was observed in the treated mice. (Wang et al., 2014; PMID: 24778279) FAP+ cells were identified using flow cytometry of lung tissue using the antibody (Kimura et al., 2019; PMID: 31188013). This antibody was used for detection of murine FAP expression on HT1080 cells by flow cytometry (de Sostoa et al. 2019, PMID: 30683154). |
