Immunogen | The original antibody was raised by immunizing a Fas-deficient mice with partially purified human Fas antigen. |
Application Notes | The original antibody was used for flow cytometry on activated T cells from humans, chimpanzees, Japanese monkeys, crab-eating monkeys and marmosets, and on thymocytes from BALB/c mice. Here it was determined that this antibody was reactive to humans and mice. This antibody was used for a cytotoxic assay on synovial cells (Ichikawa et al., 2000; PMID:10744657). The structure of the original antibody was determined using x-ray crystallography (Ito et al., 2002; PMID:11754745, Haruyama et al., 2002; PMID:12499636). This antibody was used for a competitive ELISA on Fas (Haruyama et al., 2002; PMID:12499636, Yoshida et al., 2010; PMID:20024619). This antibody was used for surface plasmon resonance on mouse Fas, to determine binding affinity. The protective effects of this antibody against acute and lethal hepatic injury induced by Jo2 was assessed. It showed it was able to protect against hepatic injury. To assess the effect of this antibody on caspases in vivo, mice were injected with the antibody after Jo2-induced apoptosis of hepatocytes. It showed that it strongly inhibited activation of caspases (Yoshida et al., 2010; PMID:20024619). |

Note on publication | In the original paper they report on the interesting characteristics of the newly obtained anti-Fas mAb, HFE7A, which cross-reacts with the Fas molecules of various species ranging from human to mouse and mitigates autoimmune symptoms without hepatotoxicity in mice. |
