Specificity | This antibody specifically reacts with the HLA-DR antigen and does not cross-react with HLA-DP or HLA-DQ. It binds to a conformational epitope on HLA-DRα, which requires the proper folding of the αβ heterodimer. HLA-DR, a major histocompatibility complex (MHC) class II cell surface receptor, is encoded by the human leukocyte antigen complex located on chromosome 6 at region 6p21.31. HLA-DR plays a key role in autoimmune conditions, influencing both disease susceptibility and resistance. Its primary function is to present peptide antigens, including those of foreign origin, to the immune system. This process helps elicit or suppress T-helper cell responses, which eventually lead to the production of antibodies against the specific peptide antigen. |

Application Notes | This antibody can be used for the immunoprecipitation of HLA-DR and MHC class-II peptides (Bonnefoy et al., 1988; PMID: 2961843). This antibody selectively inhibited the in vitro proliferation of Epstein-Barr virus-transformed lymphoblastoid cell lines (Kabelitz and Janssen, 1989; PMID: 2467753). This antibody can also be used for the immunocytochemical and immunohistochemical analysis of frozen sections of necropsy brain tissue to identify HLA-DR-expressing cells (Esiri et al., 1991; PMID: 1864982). This antibody was used in the detection of MHC class II protein in a western blot (Esser et al., 2001; PMID: 11390619). This antibody was used in a flow cytometric assay to detect the decrease in HLA-DR expression on monocytes, which is an indicator of immunosuppression in patients with sepsis and is correlated with an increased risk of secondary infection and mortality (Quadrini et al., 2021; PMID: 33432735). |
