Specificity | This antibody is specific for the human insulin-like growth factor 1 receptor (IGF1R). It recognises the extracellular portion of the receptor, specifically a conformation-dependent epitope in the alpha subunit (amino acids 62-184). It does not cross-react with rat or mouse IGF1R or human insulin receptors in co-precipitation or immunodepletion assays. |

Immunogen | This antibody was raised by immunising BALB/c mice with transfected mouse fibroblasts expressing high levels of human IGF1R (IGF1R/3T3). |
Application Notes | The ability of this antibody to bind human IGF1R has been confirmed in IGF1 binding assays using both IG1FR-overexpressing IGF1R/3T3 cells and soluble receptors (Soos et al, 1992). This antibody efficiently precipitates IGF1R solubilized from IGF1R/3T3 cells and receptor-IGF1 complexes (Soos et al, 1992). When immobilised, this antibody has been shown to remove 90% of all soluble IGF1R in immunodepletion experiments (Soos et al, 1992). It has also been used to capture IGF1R N-terminal fragments as part of a ligand plate binding assay (Molina et al, 2000). This antibody strongly stimulates both IGF1 and insulin binding to IGF1R/3T3 cells, and potentiates insulin stimulation of DNA synthesis (Soos et al, 1992). |
