Immunogen | The original antibody was selected from a llama-derived nanobody phage display library by phenotypic screening against human brain endothelial cells. |
Application Notes | The original antibody (llama VHH) bound specifically to HCEC, as demonstrated by an ELISA, and showed selective binding to brain endothelial cells (HCEC and HPEC). It localized to vesicular and membrane fractions, was internalized via an energy-dependent pathway without observed toxicity, and exhibited uptake into brain endothelial cells. A fusion protein version of this antibody (FC5-c-Myc-His5) demonstrated effective transmigration across an in vitro blood-brain barrier (BBB) model using HCEC monolayers, comparable to empty membranes, without altering BBB integrity. In vivo, the antibody accumulated in brain tissue after intravenous injection (Muruganandam et al., 2002; PMID: 11772942). This antibody's ability to facilitate transcytosis across the BBB via receptor-mediated pathways was demonstrated through significantly enhanced brain and cerebrospinal fluid (CSF) exposure in rat and dog in vivo models. |
