Immunogen | Affinity selection procedures were used to isolate this scFv from a naïve Tomlinson I phage display library that specifically binds to TNF-α.§ The panning procedure was carried out by incubating immunotubes coated with recombinant TNF-α protein with a phage suspension. Phage particles with affinity for recombinant TNF-α were amplified in TG-1 bacteria, and three rounds of panning were performed to select high affinity and specificity scFvs. |
Application Notes | The binding affinity of this scFv for human TNF-α has been confirmed in ELISA analysis (Safarpour et al, 2018). This scFv could have future therapeutic applications as a TNF inhibitor. TNF inhibition is a strategy currently showing promise in the treatment of a range of inflammatory diseases, including rheumatoid arthritis and Chron's disease. |
