Specificity | This antibody binds human VEGF-D and block the binding of VEGF-D to both VEGFR-2 and VEGFR-3. Peptide-based mapping and site-directed mutagenesis suggested that binding epitope of this antibody includes the five residues 147NEESL151of the VEGF-D (PMID: 21315726). VEGF-D signaling through the VEGFR-3 receptor has been shown to be the primary source of lymphangiogenic activation and has also been noted in pathogenic angiogenesis in some tumors. |

Immunogen | The original antibody was generated by immunizing female BALB/c mice with truncated portion of human VEGF-D from residue 93 to 201, which had FLAG-tag at its N-terminus. |
Application Notes | The original mouse antibody VD1 competes with VEGF-D to bind both VEGFR-2 and VEGFR-3 . VD1 blocks the mitogenic response of human microvascular endothelial cells to VEGF-D. The binding specificity of the original mouse version of this antibody towards human VEGF-D was determined using ELISA. The original mouse antibody was capable of immunoprecipitating human VEGF-D. This antibody was also capable of detecting VEGF-D in a western blot assay (PMID: 10785369). This antibody was used in a study involving a mouse tumor model to explore the role of VEGF-D in promoting both angiogenesis and lymphangiogenesis in tumors (PMID: 11175849). This antibody was also used to identify the role of VEGF-D in promoting tumor growth and spread though lymphatic system in a mouse model of hepatocellular carcinoma (PMID: 18338756). |
