Application Notes | The antibody is a relatively low-affinity germline-encoded antibody with a true Ka of 2.5*10^5 M-1 (Rothstein et al., 1983; PMID: 6843553). The specificity of the IgG1 format of the antibody was confirmed by ELISA analysis. The antibody was employed in in vitro binding assay, the antibody bound effectively to high density antigen beads with retention of 57.8% while the binding to low density target was 17.6%. The antibody behavior in vivo,using antigen-substituted beads s.c. implanted into SCID mice was examined. Results showed the antibody bound effectively to the HD antigen target (Zuckier et al., 2000; PMID: 11156404). The crystal structure of the Fab fragment was determined and the antibody affinity for the hapten was measured (Kd= 8.1 mM) (Sethi et al., 2006; PMID: 16618601). |
