Immunogen | The original antibody was generated by immunizing female white leghorn chickens with 50 µg of recombinant B7-H4 protein (Phe29 to Ala258) through intramuscular injection. The spleens of the chickens were harvested 7 days after the final immunization to construct an scFv antibody library. The antibody was isolated by panning against B7-H4 protein. |
Application Notes | This antibody recognized intact wild type human B7-H4 and linear epitope comprising amino acids 'VIQWLKEGVLGLVHEFK' in an ELISA. This antibody was also capable or recognizing B7-H4 in a western blot assay. The binding of this antibody to B7-H4 protein can effectively neutralize the immunosuppression response induced by B7-H4 proteins, thus enabling T-cells to retain their activation status (PMID: 33716253). |
