Species Reactivity | Pig (Sus scrofa) |
Basic Information | |
Specificity | This antibody binds the active site crevice of the porcine pancreatic alpha-amylase and is a strong amylase inhibitor. |
Alternative Name | PA; AMY2; 1,4-alpha-D-glucan glucanohydrolase |
UniProt | P00690 |
Immunogen | The original antibody was generated by immunizing dromedary with the enzyme and isolating specific binders from the VHH cloned from peripheral blood lymphocytes. |
Application Notes | This antibody can bind porcine pancreatic alpha-amylase in an ELISA and inibits the enzyme with an IC50 = 10nM (PMID: 9649422). This antibody interacts with the amino acid residues in the alpha amylase catalytic site and has a strong inhibitary action on the catalytic activity of the enzyme. The inhibitory interaction of AMD9 involved mainly its complementarity-determining region (CDR) 2 loop, whereas the CDR3 loop was small and certainly did not protrude (PMID: 11960990). |
