3S193 (IgG3) was generated in BALB/c mice by immunisation with Ley-expressing cells of the MCF-7 breast carcinoma cell-line.
Application Notes
The murine 3S193 showed high specificity for Ley in ELISA tests with synthetic Ley and Ley-containing glycoproteins and glycolipids and also reacted strongly in rosetting assays and cytotoxic tests with Ley-expressing cells (Scott et al, 2000). Murine 3S193 was humanised and in antibody-dependent cell-mediated cytotoxicity assays displayed 100-fold greater activity (Kitamura et al, 1994). 3S193 has been used to screen tumour samples for Ley expression using immunohistochemistry (Krug et al, 2007). A crystal structure of the humanised antibody has been obtained to 1.9 Å resolution (Ramsland et al, 2004).
