Specificity
This antibody binds the extracellular domain of the human PSMA. Prostate-specific membrane antigen (PSMA) is a type II integral membrane glycoprotein belonging to the M28 peptidase family. It acts as a glutamate carboxypeptidase on different alternative substrate. It is highly expressed in cancerous cells and is used as an effective diagnostic and prognostic indicator of prostate cancer.

Immunogen
The original antibody was generated by immunizing BALB/c mice with LNCaP cells or a primary culture of PCa epithelial cells.
Application Notes
This antibody was used in the immunohistochemical analysis of PSMA expressed on renal, urothelial, colon, lung, breast carcinomas and metastatic adenocarcinoma of liver. This antibody can also immunoprecipitate PSMA from crude LNCaP lysates (PMID: 9288760). This antibody was also used for the identification of PSMA in a western blot assay. The epitope analysis of this antibody was done using ELISA. This antibody was also used in the immunofluorescence staining of on nonpermeabilized and permeabilized LNCaP cells (PMID: 9288760). This antibody was used in a flow cytometric study for enumeration of circulating prostate microparticles (PMPs), a type of extracellular vesicle (EV) released by prostate cancer present in seminal fluid, urine, and blood may represent a non-invasive means to identify and prioritize patients with intermediate and high risk of prostate cancer (PMID: 26814433). In a phase 1 trial in patients with advanced solid tumor malignancies this antibody showed acceptable toxicity and excellent targeting of known sites of metastases (PMID: 17290063).
