Immunogen | A phage display library of human single chain variable fragment (scFv) antibodies was constructed from the B cells of healthy volunteers boosted by the VACV vaccine. Human scFv was isolated by panning against VACV. |
Application Notes | The scFv fragment was converted to human IgG1 (VC212). The binding affinity of VC212 was estimated by competition ELISA to IC50 values 22 µg/ml (146.67 nM). The humanized version of the antibody neutralized the VACV, as evidenced by the plaque reduction neutralization test result with a PRNT50 of ~0.16 mg/ml (~1.07 µM) (Shin et al., 2019; PMID: 30856707). |
