Immunogen | The original antibody was raised by immunizing Fn14-deficient mice with CHO cells expressing human surface Fn14 and boosted with Fn14-myc-His |
Application Notes | The original antibody was tested to inhibit distinct TWEAK-induced cellular effects known to be mediated by Fn14. HT29 and WiDr cells were treated with this antibody, this resulted in a strong inhibition of TWEAK-induced IL8 production. Furthermore, it was shown that oligomerization can greatly increase the agonistic activity of this antibody (Salzmann et al., 2013; PMID:23532848). This antibody was used for flow cytometry on HEK293 cells transfected with plasmids expressing Fn14 protein. This antibody was used for a western blot on lysates from HEK293 cells expressing Fn14 (Trebing et al., 2014; PMID:24135629). This antibody was used for immunohistochemistry on different human brain tumors (Tran et al., 2006; PMID:17018610), and on samples from xenografted tumors made from H460 cells (Jandova et al., 2015; PMID:25563366). |
Antibody First Published | Tran et al. Increased fibroblast growth factor-inducible 14 expression levels promote glioma cell invasion via Rac1 and nuclear factor-kappaB and correlate with poor patient outcome Cancer Res. 2006 Oct 1;66(19):9535-42. PMID:17018610 |
Note on publication | The original paper shows that the fibroblast growth factor-inducible 14 (Fn14), a member of the tumor necrosis factor (TNF) receptor superfamily, is expressed at high levels in migrating glioma cells in vitro and invading glioma cells in vivo. |
