Specificity
This antibody is specific for the F1 capsule protein from Yersinia pestis. It shows some cross- reaction with other Yersinia species that have a F1 protein. F1 is a protein that is part of the capsule of Yersinia pestis. Yersinia pestis is the bacteria that causes the plague in humans.

This antibody was generated by panning a phage library prepared from 40 different samples of human peripheral blood lymphocytes against the F1 protein.
Application Notes
To determine the ability of the scFv version of this antibody to bind the F1 protein, an ELISA was performed on F1 using the scFv version of this antibody, it showed strong binding. Furthermore, an ELISA was also performed on Yersinia pestis to assess the binding in vivo. Additionally, flow cytometry was performed on both Yersinia pestis and the F1 antigen, it showed a positive reaction to both (Lillo et al, 2011; pmid:22174746).

Phage antibody panning was performed against F1 in an automated fashion using the Kingfisher magnetic bead system. Selected scFvs were screened for F1-binding specificity by one-step alkaline phosphatase enzyme linked immunosorbant assay (ELISA), and assayed for binding to recombinant antigen and/or Y. pestis by flow cytometry and whole-cell ELISA.
