Specificity
This antibody binds human hepcidin-25 near the N-terminus region and does not cross react with human hepcidin-20 or human hepcidin-22. This antibody also does not cross react with mouse or rat hepcidin-25. Hepcidin is a an iron-regulating peptide hormone made in the liver. Hepcidin is a cationic, cysteine-rich and tightly folded peptide stabilized by 4 disulfide bonds that plays a major role in innate immunity and iron homeostasis. It controls the delivery of iron to blood plasma from intestinal cells absorbing iron, from erythrocyte-recycling macrophages, and from iron-storing hepatocytes. Hepcidin acts by binding to and inactivating the sole cellular iron exporter, ferroprotein, which delivers iron to plasma from all iron-transporting cells.

Immunogen
The original antibody was generated by immunizing mice with immunized with a N-terminal hepcidin peptide 'DTHFPIC' conjugated with a five amino acid peptide linker (i.e., GPGPG) to an OVA peptide (amino acids 323-336) sequence, i.e., full-length immunogen DTHFPICGPGPGISQAVHAAHAEINE.
Application Notes
The original mouse IgG1 antibody binds human hepicidin-25 with a binding affinity of kd= 1.2 nM. The binding characterization of this antibody towards human hepcidin was done using ELISA (US8609817).
