Immunogen
The antibody was generated immunizing Llama. A phage display library was constructed from immune tissue. The antibody was selected by panning against the antigen.
Application Notes
The specificity of the original format of the antibody was confirmed by ELISA analysis (IC50 value= 15 nM). The binding affinity of the nanobody to IL-12RB2 was measured by surface plasmon resonance (Kd= 6.9 nM). The antibody recognised T cells expressing IL-12RB2, as shown by FACS analysis (EC50= 1.5 nM). The antibody could inhibit the IL-12 function and the dependent IFN-γ secretion was measured. (IC50 of IFNγ release T cells D839 = 67 nM) (US20220242958).
