Specificity
This antibody binds an epitope involving amino acids 8-10 of the A chain of human insulin and binds both human insulin and proinsulin with the same affinity. This antibody cannot recognize insulin when the chain A and chain B are dissociated. This antibody also reacts with porcine and rodent insulin. Insulin is a peptide hormone produced by the beta cells of the pancreatic islets. It reduces blood glucose concentration by promoting its absorption from the blood into liver, fat and skeletal muscle cells. Decreased or absent insulin activity results in diabetes mellitus, a condition of high blood sugar level (hyperglycemia).

Immunogen
The original antibody was generated by immunizing female BALB/c mice with human insulin in Freund's complete adjuvant.
Application Notes
The scFv version of this antibody bound insulin at an IC50 of 3.5 x 10(-8) M whereas the parent IgG1 antibody bound insulin at 1.0 x 10(-8) M (PMID: 8047074). This antibody can bind human insulin and proinsulin in an ELISA (PMID: 3894122). This antibody can also be used for the immunohistochemical staining of insulin to identify β-cells (PMID: 19571666; 31380261). This antibody was also used in double immunofluorescence co-localization experiments on the same slides of human pancreas and gastrointestinal tissue (PMID: 16161007).
