Specificity
This antibody binds human p16, which is located in the nucleus and cytoplasm. It acts as a negative regulator of the proliferation of normal cells by interacting strongly with CDK4 and CDK6. This inhibits their ability to interact with cyclins D and to phosphorylate the retinoblastoma protein.

Immunogen
The original antibody was generated by immunizing BALB/c mice with recombinant p16 antigen protein.
Application Notes
The original IgG1 version of this antibody binds human p16 protein with a affinity constant of 1.92 X 10^9 M^(-1). The binding specificity and affinity of this antibody for p16 was determined by ELISA. Western blotting experiments suggested that this antibody is capable of recognizing p16 and tumor cells expressing p16 molecules like Hela cells. The immunohistochemical staining pattern of this antibody is highly similar to another anti-p16 antibody E6H4 (CN105001327).
