Application Notes
The original version of this antibody (rat IgG2b) was used to investigate PD-1 signaling pathways in the context of allergic sensitization and challenge in mice. Specifically, it was used to determine how PD-1 blockade affects airway reactivity and immune responses (Matsumoto et al., 2004; PMID: 14764726). The original antibody was used successfully for blocking the PD-1 pathway in combination with ex vivo TKD/IL-2-stimulated NK cells in preclinical models of glioblastoma and lung cancer. It was found that this combination therapy significantly enhanced the cytotoxic activity of NK cells compared to monotherapy. The treatment resulted in improved overall survival, massive infiltration of tumors by CD8+ cytotoxic lymphocytes and NK1.1 cells, and a reduction of PD-1+ immune cells in the tumor microenvironment.

The original publication investigates the role of B7-DC in regulating asthmatic responses through an IFN-gamma-dependent mechanism, independent of PD-1 signaling, in a murine model of OVA-induced allergic asthma.
