Specificity
This antibody specifically targets human TPO, a cell surface enzyme critical for thyroid hormone synthesis. TPO is found in nearly all patients with active autoimmune thyroid disease.

A cDNA library was constructed from the thyroid tissue of patients with Grave's disease, and antibody fragments were expressed in XL1-Blue cells.
Application Notes
The original form of this antibody (human IgG1) binds human TPO with a Kd of 0.29 nM (Chazenbalk et al., 1993; PMID: 7686925). Competition assays revealed that this antibody binds a distinct epitope compared to clone TR1.9 (Chazenbalk et al., 1993; PMID: 7686925). Further competition studies demonstrated that this antibody inhibits TPO autoantibodies in patients' sera by interacting with serum TPO autoantibodies (Chazenbalk et al., 1993; PMID: 7505290). Alongside clones TR1.9, SP1.4, and TR1.8, this antibody maps to the immunodominant region of TPO recognized by autoantibodies in patient sera (Nishikawa et al., 1994; PMID: 7512572). This antibody has also been employed in flow cytometry to detect cells expressing surface TPO (Jaume et al., 1999; PMID: 10323395).
