The original antibody was generated by immunizing llamas with a cocktail of murine and human Toll-like receptors (TLRs) that contained recombinant human TLR4/MD2, recombinant human TLR3, recombinant murine TLR3, and recombinant murine TLR2/Fc chimera. Peripheral blood mononuclear cells were isolated from the animals' blood for the construction of a phage display library, and the library was panned against the TLRs used for immunization.
Application Notes
The binding specificity of the original version of this antibody (VHH) was tested using ELISA. In a cell-based neutralizing assay with hTLR3-overexpressing HEK cells, an antagonistic effect was observed, as measured by NFκB expression after cell lysis compared to the control group (WO2009138494A2).
