Specificity
This antibody binds human VEGF-D and block the binding of VEGF-D to both VEGFR-2 and VEGFR-3. Peptide-based mapping and site-directed mutagenesis suggested that binding epitope of this antibody includes the five residues 147NEESL151of the VEGF-D (PMID: 21315726). VEGF-D signaling through the VEGFR-3 receptor has been shown to be the primary source of lymphangiogenic activation and has also been noted in pathogenic angiogenesis in some tumors.

Immunogen
The original antibody was generated by immunizing female BALB/c mice with truncated portion of human VEGF-D from residue 93 to 201, which had FLAG-tag at its N-terminus.
Application Notes
The original mouse antibody VD1 competes with VEGF-D to bind both VEGFR-2 and VEGFR-3 . VD1 blocks the mitogenic response of human microvascular endothelial cells to VEGF-D. The binding specificity of the original mouse version of this antibody towards human VEGF-D was determined using ELISA. The original mouse antibody was capable of immunoprecipitating human VEGF-D. This antibody was also capable of detecting VEGF-D in a western blot assay (PMID: 10785369). This antibody was used in a study involving a mouse tumor model to explore the role of VEGF-D in promoting both angiogenesis and lymphangiogenesis in tumors (PMID: 11175849). This antibody was also used to identify the role of VEGF-D in promoting tumor growth and spread though lymphatic system in a mouse model of hepatocellular carcinoma (PMID: 18338756). This antibody was used to study the localization of VEGF-D in primary tumors by immunohistochemical analysis (PMID: 12148568).
