Immunogen | The original antibody was isolated from a human scFv phage display library by panning against human CD72 receptor using human tonsillar mononuclear cells. The phage library was further enriched using peripheral blood B lymphocytes as target in presence of anti-CD19 antibody. The final round of panning was done against peripheral blood CD19+B cells and subsequently tested for binding to CD72- transfected L929 cells. Later on the scFv antibody was converted to human IgG1 format using antibody engineering. |
Application Notes | The binding of this antibody to human CD72 expressed on the surface of Ramos Burkitt's lymphoma cells and hCD72 transfected L929 cells was determined using flow cytometry. This antibody was also used in the immunohistochemical staining of frozen sections of various human tissues expressing human CD72. Internalization assays with Ramos Burkitt's lymphoma cells were performed in order to determine the internalizing capacity of this antibody. It was reported that this antibody is capable of being internalized and are resistant to protease treatment (WO2004067569). |
