Immunogen | The original antibody was generated by immunizing DBA/1 mice with chicken or rat CII. |
Application Notes | Epitope localization of the original format of this antibody (mouse IgG2b) was conducted using ELISA (Schulte et al., 1998; PMID: 9430695). this antibody, in combination with CIIC1, was utilized to induce collagen type II (Col II) antibody-induced arthritis in vivo in mice (Nandakumar et al., 2003; PMID: 14578183). This combination has later been shown to be potently arthritogenic in vivo, affecting cartilage matrix integrity in cartilage explant cultures, impairing synthesis of new matrix, and disrupting collagen fibril formation in chondrocyte cultures (Croxford et al., 2013; PMID: 23239042). Moreover, this antibody was employed in a study to investigate whether mAb CIIF4, when given in combination with this antibody and CIIC1, can prevent cartilage damage in bovine cartilage explants in vitro (Croxford et al., 2010; PMID: 20662051). The original version of this antibody was utilized to examine the production of collagen II with IHC staining (Gharravi et al., 2016; PMID: 26254592) and in the immunohistochemical staining of Col II (Soetjahjo et al., 2018; PMID: n/a). |
Antibody First Published | Schulte et al. Arthritis-related B cell epitopes in collagen II are conformation- dependent and sterically privileged in accessible sites of cartilage collagen fibrils J Biol Chem. 1998 Jan 16;273(3):1551-61. PMID:9430695 |
Note on publication | The original publication investigates the development of antibodies against collagen type II (CII) in the context of arthritis, focusing on the characterization of arthritis-related conformational epitopes and highlighting the influence of the triple-helical conformation on antigen recognition, alongside a preimmune B cell selection process biased by the accessibility of CII determinants in intact cartilage tissue. |
