Specificity | This antibody is specific for cortactin in both birds and mammals. It recognizes 80 kDa and 85 kDa Cortactin in avian cells, and also recognizes 80 kDa protein in rodent and human cells. Cortactin contributes to the organization of the actin cytoskeleton and cell shape. |

Immunogen | The mouse version of this antibody was generated by immunizing mice with a mixture of affinity-purified tyrosine phosphoproteins from chick embryo fibroblasts expressing activated pp60src, and recognizes Cortactin. |
Application Notes | To characterise the binding of the mouse version of this protein to tyrosine- phosphorylated protein, immunoblotting on tyrosine-phosphorylated proteins from CEFs, NIH 3T3, Rat-1, HOS and 10W was performed. Furthermore, immunoprecipitation on the same samples was preformed. The antibody showed a reaction in both experiments and had cross-reactivity with the other species (Kanner et al, 1990; pmid:2110361). To confirm the specificity of the mouse version of this antibody, immunoprecipitation of cortactin was preformed using this antibody. Furthermore, the immunoprecipitated antibodies were western blotted with this antibody. The western blot showed bands corresponding to cortactin. Finally, invadopodia were immunostained for cortactin using this antibody and showed co- localization with antibody clone #170-10, which is also an anti-cortactin antibody (Bowden et al, 1999; pmid:10442635). |

Note on publication | To study transformation-relevant tyrosine kinase substrates, we have generated monoclonal antibodies to individual tyrosine phosphoproteins, including p130, p120, p110, and five additional phosphoproteins (p210, p125, p118, p85, and p185/p64). These antibodies detected several of the same tyrosine phosphoproteins in chicken embryo fibroblasts transformed by avian retroviruses Y73 and CT10, encoding the yes and crk oncogenes, respectively. |
