Basic Information | |
Specificity | This antibody binds cortisol. It has also shown cross-reactivity to dexamethasone and prednisolone. This is due to a hydroxyl group bound to C11. Cortisol is a steroid hormone, in the glucocorticoid class of hormones. It is produced in many animals, mainly by the zona fasciculata of the adrenal cortex in the adrenal gland. It is produced in other tissues in lower quantities. It is released with a diurnal cycle and its release is increased in response to stress and low blood-glucose concentration. It functions to increase blood sugar through gluconeogenesis, to suppress the immune system, and to aid in the metabolism of fat, protein, and carbohydrates. It also decreases bone formation. This antibody has not shown any affinity for cortisol-21-hemisuccinate-BSA. |

Immunogen | The original antibody was generated by immunizing mice with cortisol-7-CM, followed by cortisol-3-CMO. Both forms of cortisol were bound to BSA. |
Application Notes | The specificity and affinity of this mouse version of this antibody was determined using equilibrium dialysis in dialysis cells fitted with a cellulose membrane of 3500 MW cutoff (Calvez et al, 1995; pmid:7898495). Further a radioimmunoassay was performed on I125-cortisol using the scFv version of this antibody (Calvez et al, 1995; pmid:7898495). The specificity of this antibody towards cortisol-3-CMO-BSA, cortisol, prednisolone and dexamethasone was also tested using equilibrium dialysis. ELISA was used to show that this clone was able to bind steroids conjugated to BSA through nonspecific hydrophobic interactions (PMID: 9820517). |
