Specificity | This antibody is specific for human CXCR3, a receptor for the C-X-C chemokines CXCL9, CXCL10, and CXCL11. CXCR3 mediates the proliferation, survival, and angiogenic activity of human mesangial cells (HMC) via a heterotrimeric G-protein signaling pathway. The CXCR3 isoform 2 also regulates the proliferation, survival, and angiogenesis of microvascular endothelial cells (HMVEC) through a cAMP- mediated signaling mechanism. Dysregulation of CXCR3 signaling has been linked to several disorders, including type 1 diabetes mellitus and psoriasis. |

Application Notes | This human IgG1 antibody was evaluated for its effector functions, including Antibody- Dependent Cell-Mediated Cytotoxicity (ADCC) assays, where it was tested with primary human NK cells and the NK9.2 cell line as effector cells against CHO cells expressing human CXCR3. The antibody demonstrated cytotoxicity against CXCR3-expressing target cells. Binding studies using a Biacore T200 revealed that 53Hu37 displayed differential binding affinities to human and mouse Fcγ receptors, which are important for ADCC and complement-dependent cytotoxicity (CDC). In vivo, the antibody was administered to cynomolgus monkeys, leading to a reduction in effector memory CD4+ and CD8+ T-cells, as well as CXCR3 staining in the spleen. Biochemical stability analyses showed that 53Hu37 maintained favorable stability under various stress conditions. |
