Specificity | This antibody is specific for the H1N1 influenza virus hemagglutinin protein. It recognizses a conserved non-linear epitope (residues 133, 137 and 222) on the globular head of H1N1 HA between the receptor-binding pocket and the Ca2 antigenic site. The antibody inhibits all tested H1N1 influenza strains from 1918 to 1977 (A/South Carolina/1/1918, A/swine/Iowa/15/1930, A/Weiss/1943, A/L3/1947, A/New Jersey/11/1976, A/USSR/92/1977) and the pandemic 2009 virus but not the seasonal H1N1 strains from 1999 or 2007. |

Immunogen | The original antibody was isolated from the peripheral blood mononuclear cells of a healthy 47-year-old human subject. |
Application Notes | The crystal structure of the Fab fragment in complex with a bacterially expressed and refolded globular head domain from the hemagglutinin was determined (Hong et al., 2013; PMID: 24027321). The antibody was inhibitory in a hemagglutination inhibition activity against H1N1pdm2009-like virus. Neutralization capacity of the antibody was prooved by an MN-assay. The specificity of the antibody to the monomeric head as to the full HA-trimer was confirmed by ELISA (Aartse et al., 2021; PMID: 34358138). |
