Application Notes | Lysozyme is an enzyme that degrades polysacchardies of various types of bacterial cell walls. It is observed that lysozyme bound D1.3 still retains its enzymatic activity. This is attributed to the fact that D1.3 binds lysozyme at a site remote from the catalytic site and that there is no significant conformational change caused by the binding (PMID: 3614217). The three dimension structure of a complex between the Fab from antibody D1.3 and its antigen, HEL, has been determined at 6 and at 2.8 a resolution (PMID: 2426778, 3965976). Sixteen residues of lysozyme make contact with 17 residues of the antibody. The antigenic determinant is topographic including lysozyme residues 18, 19, 21–25, 27, 116–121, 124, and 129. All six complementarity determining regions of the antibody contact the antigen, and in addition, two residues from “framework” segments of V H and V L (Amit et al., 1986; PMID: 2441250). D1.3 binds native lysozyme in an ELISA (PMID: 2448386). |
