Immunogen | The original antibody was generated by immunization of Llama glama with mCherry fluorescent protein. A VHH cDNA library was generated by high-throughput DNA sequencing of a marrow lymphocyte from an immunized llama. Later on this was combined with mass spectrometric (MS) identification of high affinity VHH regions derived from serum of the same animal. |
Application Notes | This antibody can be used to detect mCherry fluorescent protein and DsRed in an ELISA. This antibody binds immobilized mCherry with a binding affinity of Kd= 0.18 nM. The Alexa Fluor® 488 conjugated version of this antibody was used to immunofluorescently stain S. cerevisiae expressing mCherry-tagged histone H2B (PMID: 25362362). This antibody was used for the visualization and manipulation of red fluorescent labelled proteins in live cells by using nanobodies targeting fluorescent protein tags (PMID: 31122229). |
