Immunogen | This antibody was raised by immunizing a BALB/c mouse with human osteopontin. Then the antibody was humanized. |
Application Notes | The original antibody was used for an ELISA on human osteopontin to determine its specificity. The mouse version of this antibody was used for a western blot on both human and mouse osteopontin, here it was shown that the antibody reacts to both human and mouse osteopontin. This antibody was tested in vivo on CIA mice spleens to test the effect on lymphocytes migration, it showed that the antibody could effectively block lymphocyte migration. Furthermore the antibody could also effectively promote the apoptosis of activated lymphocytes (US20100239588A1, Fan et al., 2008; PMID:18576331). The mouse version of this antibody was used in vitro to determine its effect on ovariectomy-induced osteoporosis, in these experiments it showed that it was able to show significant inhibition in bone loss, further it showed that it was able to reduce mature OC mediated- bone resorption, finally it promotes OC apoptosis (Zhang et al., 2014; PMID:25201732). |
Antibody First Published | Fan et al. Treatment of collagen-induced arthritis with an anti-osteopontin monoclonal antibody through promotion of apoptosis of both murine and human activated T cells Arthritis Rheum. 2008 Jul;58(7):2041-52. PMID:18576331 |
Note on publication | In the original paper they tested the effects of a novel monoclonal antibody (mAb) against human osteopontin (OPN) in the prevention and treatment of collagen-induced arthritis (CIA) and to elucidate the underlying mechanisms of these effects. |
