Specificity | This antibody binds to a conformational epitope on human PAI-1 which comprises the loop connecting strands 1 and 2 of β-sheet B (s1B and s2B) as well as the proximal portions of each strand. Plasminogen activator inhibitor-1 (PAI-1), is a serine protease inhibitor, a unique member of the serpin superfamily which plays an important role in fibrinolysis and is an established risk factor for cardiovascular diseases. It a primary inhibitor of tissue-type plasminogen activator (PLAT) and urokinase-type plasminogen activator (PLAU). As PLAT inhibitor, it is required for fibrinolysis down-regulation and is responsible for the controlled degradation of blood clots. As PLAU inhibitor, it is involved in the regulation of cell adhesion and spreading . |

Application Notes | This antibody is reported to exhibit moderate dose-dependent PAI-1 inhibition. This antibody is capable of inhibiting PAI-1 activity by directly preventing the initial PAI-1.t-PA complex formation (PMID: 27604413). This antibody interferes with exosite interactions between 37-loop of proteinase and negatively charges residues of the RCL and affects the formation of Michaelis complex and dissociation of acyl enzyme (Komissarov et al, 2019). |
