| Specificity | This antibody binds human PAI-1 in the hI‐s5A loop comprising amino acid residues 300 to 314 (RQFQADFTSLSDQ). Plasminogen activator inhibitor-1 (PAI-1), is a serine protease inhibitor, a unique member of the serpin superfamily which plays an important role in fibrinolysis and is an established risk factor for cardiovascular diseases. It a primary inhibitor of tissue-type plasminogen activator (PLAT) and urokinase-type plasminogen activator (PLAU). As PLAT inhibitor, it is required for fibrinolysis down-regulation and is responsible for the controlled degradation of blood clots. As PLAU inhibitor, it is involved in the regulation of cell adhesion and spreading . |

| Immunogen | The original antibody was isolated from a nanobody library generated by immunization of three alpacas with three different PAI-1 variants. | 
| Application Notes | This antibody is capable of inhibiting both glycosylated and non-glycosylated PAI-1 variants through a substrate-inducing mechanism. The profibrinolytic effect of this antibody was confirmed using an in vitro clot lysis assay (PMID: 27604413). This antibody neutralized PAI-1 by redirecting (95-97%) of the reaction with two chain (tc) tPA and uPA plasminogen activators to the substrate branch. This antibody was less effective with single chain proenzymes (Komissarov et al, 2019). |
