Application Notes | This stable, recombinant single-chain Fv-Cκ (scFv-Cκ) antibody fragment was generated by protoplast fusion of J558L myeloma cells with Escherichia coli K803 transformed with an appropriate scFv-Cκ expression construct. The aptly named M55 scFv-Cκ expression construct — containing a CDR2 Ser55-to-Arg substitution (T-to-G transition at the third base), found in the VH chain of several hypermutated antibodies (VI22, VI43, and VI53) — was derived from the secondary antibody clone VI24. M55 binds a defined subsite within the major antigenic site of VSV-G. However, the exact binding site in the subunit is not defined. M55 binding to VSV-G was measured using solid-phase ELISA. The VH Ser55-to-Arg substitution in M55, which is not found in its parental clone VI24, increased monovalent binding by about 10-fold compared with M0 (the germline derived scFv-Cκ). |
