Immunogen | The original antibody was generated by immunizing BALB/c mice with second extracellular loop of the C3aR (amino acid residues 173–332) fused to glutathione-S-transferase (GST). A combinatorial phage library was generated expressing scFv antibodies and panned against second extracellular loop of the C3aR, fused to the maltose binding protein as antigen. |
Application Notes | The original scFv antibody binds to the 2e loop of the human C3a receptor in an ELISA. Human C3a receptor is a G protein-coupled receptor stimulating chemotaxis, superoxide anion generation and granule enzyme release. This antibody was able to bind RBL-2H3 cells stably expressing the C3aR and to the human mast cell line HMC-1 in a flow cytometric assay. This antibody is not capable of blocking the interaction between C3a and C3aR (PMID: 11373092). |
