Specificity | This antibody recognizes the protruding (P) domain of the Norwalk virus capsid protein VP1 with high affinity. Specifically, it binds the P1 subdomain (located in the lower region of the P domain), which is occluded on intact particles. Still, this antibody binds intact norovirus VLPs and virions. This antibody has been shown to bind to the GII.10, GII.1, GII.2, GII.4, GII.12, GII.17 and GI.11 genotypes. |

Immunogen | The original antibody was generated by immunizing a single alpaca with the GII.10 virus-like particle protein. |
Application Notes | The reactivity of the original antibody (alpaca VHH) against norovirus VLPs and P domains was determined using ELISA. It was shown to be reactive with antigenically distinct norovirus VLPs,including GII.4, GII.10, and GII.12. Using isothermal titration calorimetry, a Kd of 7 nM was measured against GII.10 P domain and a Kd of 3.5 nM against GII.4 P domain. The crystal structure of this antibody in complex with norovirus GII.10 P domain, Saga-2006 GII.4 P domain, and 2012 NSW GII.4 P domain were determined. This antibody can bind intact norovirus VLPs and virions and promotes their disassembly (Koromyslova & Hansman, 2015; PMID: 25520510). The epitope of this antibody was further characterized in X-ray crystallography and cryo-EM. This antibody's binding mechanism involves impairing capsid motions and integrity, which inhibits multiple stages of the Norovirus life cycle (Koromyslova & Hansman, 2017; PMID: 29095961). |
