Application Notes | The specificity of this antibody's original format (human scFv) was validated via ELISA, demonstrating its selective binding to CA IX-expressing cells. It was further confirmed through flow cytometry and cross-competition studies. This antibody exhibited an IC50 of 2.086 µg/ml / 2.9 nM to stable G250-expressing 293T cells. This antibody was tested in vivo in Ncr Nude mice (athymic mice); it was incorporated into lentiviral vectors to create anti-CA IX chimeric immune receptors for T-lymphocyte transduction, resulting in enhanced T-lymphocyte cytotoxicity against renal cell carcinoma (RCC) cells. An scFv-Fc fusion version of this antibody was successfully used in an internalization study of anti-CA IX scFv- Fc antibody fusion proteins into stable G250-293T cells, exhibiting mild (~20%) inhibition (US8466263B2). |
