Immunogen | The original antibody was generated by immunizing a male lewis rat with semi-purified recombinant mammalian-expressed IL-5. |
Application Notes | This antibody can effectively block IL-5 induced HL-60 histamine content and TF-1 proliferation. 39D10 was also evaluated for its ability to neutralize the ILJ-dependent growth of eosinophilbasophil colonies (i.e., both histamine-positive and histaminenegative Eo-type) in methylcellulose culture. This antibody did not cross-react with mouse IL-5 in indirect ELISA (PMID: 1706953). This antibody was used in immunohistochemical method to study the localisation and phenotype of individual cytokine producing cells in synovial biopsy specimens in rheumatoid arthritis (PMID:7677442). This antibody was also used in immunocytochemical and a flow cytofluorimetric method to detect intracellular IFN-g, IL-4 and IL-5 in T-cell clones, peripheral blood mononuclear cells PBMC and bronchoalveolar lavage fluid BALF cells (PMID: 9134033). The original rat IgG2a antibody binds human IL-5 with a binding affinity of Kd= 53 pM (PMID: 7613159). |
Antibody First Published | Denburg et al. Interleukin-5 is a human basophilopoietin: induction of histamine content and basophilic differentiation of HL-60 cells and of peripheral blood basophil-eosinophil progenitors. Blood (1991); 77(7):1462-8. PMID:1706953 |
Note on publication | Describes the use of this antibody to study the effect of recombinant human interleukin-5 on myeloid colony formation in methylcellulose and on alkaline passaged HL-60 basophilic cell differentiation. |
