Introduction
Procalcitonin (PCT) is a serum biomarker of bacterial infections and sepsis. In dogs with infections, we found levels up to 150 ng/ml.
Principle Of The Assay
The other is conjugated to HRP and used for detection. The standard is prepared from recombinant dog Procalcitonin. Standards and diluted samples (100 μl) are incubated in antiPCT coated microtiter wells for 45 minutes. After washing the wells, HRP-conjugate (100 μl) is added and incubated for 45 minutes. If PCT molecules are present, they are sandwiched between the capture and detection antibodies. The wells are then washed to remove unbound HRP-conjugate. TMB is added and incubated for 20 minutes. If PCT is present, a blue color develops. Color development is stopped after 20-minutes by addition of Stop Solution, changing the color to yellow. Absorbance is measured at 450 nm. The concentration of PCT is proportional to absorbance and is derived from a standard curve.
