Immunogen
This conformation specific synthetic antibody was generated by utilizing a phage display library wherein the the Fab fragments is fused to the minor coat protein pIII of the M13 bacteriophage. To generate sABs that preferentially bind to the closed form of MBP (closed-specific sABs), the selection was carried out in the presence of 10 μM maltose. After fours rounds of selection the isolated colonies were subjected to competitive phage ELISA to provide an estimate of affinity towards MBP and specificity for closed form.
Application Notes
Maltose-binding protein (MBP) is a part of the maltose/maltodextrin system of Escherichia coli, which is responsible for the uptake and efficient catabolism of maltodextrins. Maltose- binding protein (MBP) is one of the most popular fusion partners being used for producing recombinant proteins in bacterial cells. MBP allows one to use a simple capture affinity step on amylose-agarose columns, resulting in a protein that is often 70-90% pure. In addition to protein-isolation applications, MBP provides a high degree of translation and facilitates the proper folding and solubility of the target protein (PMID: 20978971).
